目的:采用大鼠30%Ⅲ°烫伤模型,观察:①大鼠烧伤后及不同时机切痂后血循环中和脾脏、肠系膜淋巴结内Th1型细胞因子γ干扰素(IPN-γ)、白细胞介素-2(IL-2)、Th2型细胞因子白细胞介素-4(IL-4)、白细胞介素-10(IL-10)变化的规律:②上述细胞因子mRNA在T淋巴细胞和脾脏、肠系膜淋巴结(MLN)内表达的动态改变;③烧伤大鼠血清和焦痂提取物对离体大鼠T淋巴细胞Th1/Th2型细胞因子的变化规律的影响;④相应的T淋巴细胞转化率。探讨烧伤焦痂持续存在对机体免疫功能的危害,论证休克期切痂的必要性。 方法:清洁级雄性Wistar大鼠160只,随机分为四组:A组为单纯烧伤对照组;B、C、D组均为切痂组,分别于伤后8、24、96小时一次切除焦痂移植同种异体皮封闭创面,设立烧伤前、烧伤后4、12、24、48、96、120、168小时诸时相点,各时相点n=8。在各时相点处死动物,留取血液和脾脏、肠系膜淋巴结标本并即刻提取纯化T淋巴细胞,ELISA法检测IFN-γ、IL-4,放射免疫分析法检测IL-2、IL-10蛋白水平、RT-PCR方法半定量测定IFN-γ、IL-4 mRNA表达。以烧...
