    The invention provides a two-step enzyme method used cephalosporin c to make 7-aminocephalosporanic acid. The used D-amino acid oxidase is the purified triangle yeast D-amino acid oxidase mutant whose specific activity higher 105% than parent strain D-amino acid oxidase. There is no need to add hydrogen peroxide, beta-lactamase inhibitor, catalase inhibitor, and catalase in this method. Its yield can reach over 93%. Thus it has the advantages of simple steps, low cost, and high yield.
